Separation and purification of total flavonoids from Lycium Barbarum Linnaeus by macroporous resins

Abstract

Six macroporous resins, including of NKA-9, S-8, XDA-L, AB-8, HPD-100 and HPD-600, were studied on separation and purification of total flavonoids in Lycium Barbarum Linnaeus. The experimental results showed XDA-1 resin had better adsorption and desorption performance than other resins. With XDA-1 resin, the effects of pIL extract concentration and flow rate on the adsorption performance were studied. The dynamic adsorption and desorption conditions of XDA-1 resin for total flavonoids were optimized. The optimum dynamic adsorption conditions for total flavonoids were obtained as follows: total flavonoids concentration, 0.25mg/mL; pH, 5; flow rate, 0.5mL/min. The optimum desorption conditions were obtained at eluant, 80% ethanol aqueous solution (v/v); the amount of elution, 35mL (5 bed volume). In these techniques, total flavonoids content were increased from 27.2% to 79.8%, with a total flavonoids recovery rate of 87.4%. The results indicated that XDA-1 resin could be used in separation and purification for total flavonoids in Lycium Barbarum Linnaeus.
